In my opinion, the historical Old Town and charming Lesser Town (Mala Strana) are the best neighborhoods to stay in during your first Prague visit due to their central location. You will be a stone’s throw away from the major tourist sites such as Old Town Square, Charles Bridge, and Prague Castle. I don’t see a point of staying in other areas if you can afford to stay in the historical center since you will be commuting there anyway.

Old Town, on the other hand, is perfect for a cold-weather getaway, making it an excellent choice if you’re visiting Prague in late autumn, winter, or early spring. Another key difference is that Old Town is livelier and better suited for partygoers and shopping enthusiasts, while Lesser Town is more quaint and serene, ideal for couples seeking a romantic retreat.
